What the current record supports
Qualification
Limitations and evidence gaps
The human evidence is sponsor-concentrated, and no independent human trial had replicated a clinical effect at the evidence cutoff. Complete peer-reviewed reports remain pending for several Phase 3, conference, and sponsor-disclosed findings. Publications, registry entries, and analyses from the same underlying trial are not independent datasets and must not be double-counted.
Cardiovascular and kidney outcomes, histologic liver improvement, durability after discontinuation, structural joint effects, and direct comparisons with tirzepatide or semaglutide remain unresolved. Cross-trial numerical comparisons cannot establish superior efficacy or tolerability.
Gastrointestinal adverse events, treatment discontinuation, increased heart rate, and dysesthesia have been observed, but complete Phase 3 safety tables and longer follow-up are not yet available. Rare or long-latency outcomes, immunogenicity, pregnancy, neoplasms, thyroid events, psychiatric events, retinopathy, and the clinical significance or reversibility of dysesthesia remain incompletely characterized.
No public integrated safety summary or regulator review was available at the evidence cutoff, and several completed early-phase datasets lacked public safety results. Public reproductive, developmental, genotoxicity, carcinogenicity, and other retatrutide-specific toxicology coverage is incomplete; absence of public data is not evidence of safety.
